The Effect of Pinaverium on Isolated Guinea-Pig and Rabbit Stomach Fundus Contracted With Acetylcholine and Its Relation With Calcium

In this in vitro study, by using Ca-I-2 -free me-dium, the effect of pinaverium on acetylcholine (ACh) responses induced by cumulative addition. of Ca+2 (10-4 - 10-2 M) was investigated in isolated gu-inea pig and rabbit stomach fimdus. The responses induced with 104 M ACh, Ca+2 -free medium containing 0.77 mM Na7 EDTA, sho-wed a dose-dependent and significant increase by addition of cumulative concentration of Cal-2 . In the presence of pinaverium (10-5M), the maximum response decreased significantly in both pre-parations. It was showed that the inhibition induced by pinaverium was higher in guinea-pig stomach fundus than in rabbit stomach ftmdus (P<0.05). The results suggest that extracellular Ca+2 - int-lux can be inhibited by pinaverium in both pre-parations.
