The effect of vitamine E and ursodeoxycholic acid on immune system in obstructive jaundice

Purpose: The aim of this study is to investigate the effect of Vitamine E and Ursodeoxycholic acid (UDCA) on the leucocyte count, the ratio of T, B and Null leymphocyte and translocation of E. coli labelled vvith flourescein in an experimental obstructive jaundice model in rats. Material and Methods: Seventy male VVistar-albino rats vvere used in this study. The animals vvere divided into four groups ineluding 15 rats and obstructive jaundice vvere per- formed in ali rats. İn the first group; obstructive jaundice was performed only, İn the second group; 25 mg/kg UDCA was given orally between 8-14 days. İn the third group; 500 mg/kg/im Vitamine E was given in every two days betvveen 8-14 days (three times). İn the fourth group Vitamine E and UDCA vvere given together as men- tıoned above. İn the left ten rats; sham procedure was performed (flfth group). Two mİ solution ineluding 109 E. coli labelled vvith flourescein was given via a naso-gastric tube at 14th day. Ali rats vvere sacrificed at 15th day and mesenteric lymphatic nodules (MLN), liver, spleen, small bovvel tissue samples and one mİ blood samples vvere taken. Leucocyte types vvere deteeted by May-Grüwald-Giemsa method and T, B and Null lemphocyte ratio vvere found by ANAE enzyme. Serum Bilirubine, SGOT, SGPT and Alkaline phosphatase levels vvere measured. Translocated E. coli number deteeted in bovvell wall, MLN, liver, spleen and blood samples by flourescein mic- roseopy. Findings: There was a significant decrease of lemphocyte ratio in group I and II (p<0.05). The lemp­ hocyte ratio of groups II and III was similar vvith control group. T and B lemphocyte ratio decreased in ali ja- undiced groups. The decrease vvas significant in third and fourth groups (p<0.05). Null lemphocyte ratio inereased in ali groups, but the differences vvas significant in groups III and IV (p< 0.05). E.coli translocation vvas significant in group I, and there vvas no differences in other groups. Conclusion: Vitamine E caused Null lemphocyte pro- liferation. UDCA has no direct effect on immune system, but it prevented the lebelled E.coli translocation.
